어느 QA 팀에게 물어도 ALCOA가 무엇의 약자인지는 막힘없이 답합니다. 그런데 지난달 기록 하나 — 크로마토그램, 배치기록 한 장, 환경모니터링 로그 — 를 꺼내 놓고 그 위에서 열 개 속성을 모두 증명해 보라고 하면, 회의실이 조용해집니다.

그 간극이 이번 주제의 전부입니다.

불편한 질문을 하나 던집니다. 지난달 제품을 출하하며 근거로 삼은 기록 하나를 고르십시오. 시스템 자체로부터 누가 만들었고, 언제였고, 원래 무엇이라고 적혀 있었고, 왜 바뀌었는지를 보여줄 수 있습니까?

POINT 1

열 번째 글자는 더 이상 해설이 아닙니다. Traceable(추적성) 은 그동안 업계의 관행적 표현이었습니다. 2025년 7월 EU GMP Chapter 4 개정 초안에서 이것은 규정 문언이 되었습니다. 초안 용어집은 ALCOA++ 를 "attributable, legible, contemporaneous, original and accurate"의 약자로 정의하면서 "complete · consistent · enduring · available and traceable 속성에 추가적 강조를 두는 것"이라고 규정하며, #4.63은 이 원칙들이 종이 시스템과 전자 시스템 모두에 적용된다고 명시합니다. 세 규제기관, 세 개의 약어 — 가장 엄격한 쪽에 맞춰 설계하십시오. FDA의 CGMP 데이터 완전성 가이던스는 여전히 기본 ALCOA 를 씁니다. "attributable, legible, contemporaneously recorded, original or a true copy, and accurate." 실제로 무너지는 속성은 유명한 것들이 아닙니다. Legible(판독성) 로 지적받는 곳은 없습니다. 무너지는 것은 Original(원본성) — 재처리 가능한 파일이 진짜 원본인데 출력물을 기록으로 보관하는 경우, Contemporaneous(동시성) — 이면지에 적었다가 나중에 옮겨 적는 경우, Complete(완전성) — 정당하게 무효화한 결과를 조사보고서와 함께 남기지 않고 조용히 빼버리는 경우, 그리고 Consistent(일관성) — 기기 이력과 시험기록이 서로 대조되지 않는 경우입니다.

ALCOA++는 SOP 뒤에 덧붙이는 체크리스트가 아닙니다. 어떤 기록에 대해서든 사람의 설명 없이 시스템으로부터 답할 수 있어야 하는 열 개의 질문입니다. 실사관에게 누군가 말로 설명해 주어야 답이 된다면, 그 속성을 지고 있는 것은 기록이 아니라 사람입니다.

종이라고 예외가 아닙니다. Chapter 4 개정 초안은 이 원칙을 종이와 전자에 동일하게 적용하며, 문서의 범위를 텍스트·이미지·영상·음성까지 확장합니다. 육안검사나 배지 판독의 증거로 쓰는 사진 한 장도 규제 기록이며, 언제 찍혔고 변경되지 않았음 에 대한 방어 가능한 답이 있어야 합니다.

밑바탕 원칙: ALCOA++는 데이터가 어떻게 생겨야 하는지를 말하지 않습니다. 데이터가 당신의 도움 없이 스스로에 대해 무엇을 증명할 수 있어야 하는지를 말합니다.

Topic 02 — ALCOA++: Ten letters everyone recites, few can demonstrate

Ask any QA team what ALCOA stands for and you will get a clean answer. Ask them to take one record from last month — a chromatogram, a batch record page, an environmental monitoring log — and demonstrate all ten attributes on it, and the room goes quiet.

That gap is the whole topic.

Here is the uncomfortable question: pick one record you released product on last month. Can you show, from the system itself, who made it, when, what it originally said, and why it changed?

A few points worth debating 👇

POINT 1

The tenth letter is no longer commentary. Traceable used to be an industry habit. In the July 2025 draft revision of EU GMP Chapter 4, it became regulatory text: the draft glossary defines ALCOA++ as an acronym for "attributable, legible, contemporaneous, original and accurate", which "puts additional emphasis on the attributes complete, consistent, enduring, available and traceable", and #4.63 sets out those principles as applicable to both paper and electronic systems. POINT 2

Three regulators, three acronyms — design to the strictest. FDA's CGMP data integrity guidance still uses plain ALCOA — "attributable, legible, contemporaneously recorded, original or a true copy, and accurate." (Source: FDA, Data Integrity and Compliance With Drug CGMP: Questions and Answers, December 2018) The draft Annex 11 uses ALCOA+. The draft Chapter 4 uses ALCOA++. If you supply more than one market, the question is not which acronym is correct — it is which one you build to.

POINT 3

The attributes that actually fail are not the famous ones. Nobody fails Legible. Sites fail Original (a printout kept as the record while the reprocessable file is the real original), Contemporaneous (recorded after the fact, from a scrap note), Complete (a legitimately invalidated result quietly dropped instead of retained with its investigation), and Consistent (an instrument history that does not reconcile with the test record).

POINT 4

ALCOA++ is not a checklist to append to an SOP. It is ten questions you should be able to answer about any record, from the system, without a person explaining it. If answering requires someone to walk an inspector through it verbally, the record is not carrying the attribute — the person is.

POINT 5

Paper is not exempt. The Chapter 4 draft applies the principles to paper and electronic alike, and extends documentation to text, image, video and audio. A photograph used as evidence of a visual inspection or a plate reading is a regulated record — and it needs a defensible answer for when and unaltered.

The principle underneath it all: ALCOA++ does not describe what your data should look like. It describes what your data should be able to prove about itself without your help.
